Membrane separations
Xampler laboratory membrane cartridges GE Healthcare Xampler™ ultrafiltration and microfiltration cartridges are designed for laboratory scale processing, with typical volumes ranging from a few hundred mls to about 10 liters of solution. Xampler cartridges are offered with nominal flowpath lengths of 30 and 60 cm and membrane areas ranging from 110 to 1,400 cm2 (0.12 to 1.5 ft2). They are directly scalable to GE Healthcare 30- and 60-cm pilot and process scale cartridges. Xampler cartridges have fully self-contained housings and are well-suited for use with GE Healthcare’s QuixStand™ benchtop system. When operated in a vertical orientation complete process fluid drainage and maximum product recovery is achieved. Xampler hollow fiber cartridges are offered with mini Tri-Clamp® end fittings for quick and easy sanitary connection to your laboratory equipment. These cartridges are identified with an “M” in the suffix of the model number. A select portion of the Xampler line is also offered with barbed end fittings for simple flexible tubing connection. Schematic diagrams showing the alternative cartridge sizes and end fittings are provided in Figure 4. All GE Healthcare Xampler microfiltration cartridges are autoclavable. Moreover, all Xampler ultrafiltration cartridges are autoclavable with the exception of 1,000 Dalton nominal molecular weight cutoff (NMWC) membrane units. An “A” as the model number suffix signifies that the particular cartridge may be autoclaved.
Figure 1. Xampler hollow fiber cartridges — Xampler cartridges are available in two housing diameters and two lengths, and are designed for use with QuixStand benchtop systems
Xampler cartridge features and benefits Versatile • Nominal flowpath lengths of 30 and 60 cm facilitate optimization of process conditions and assist future scale-up • Membrane areas from 110 to 1,400 cm2 (0.12 to 1.5 ft2) suit specific processing needs • UF pore sizes range from 1 kD to 750 kD NMWC • MF pore sizes range from 0.1 to 0.65 micron
Designed for the laboratory • Low flow rate requirements allow use of smaller pumps • Polysulfone membrane minimizes non-specific protein binding and provides high product recovery • Autoclavable (with the exception of 1,000 NMWC) devices address the need for small-volume sanitary processing
Easy to use • Self-contained housings with translucent polysulfone shells allow ease of installation and provide a visual observation of the membrane separation process • Offered with Tri-Clamp end fittings for quick and easy aseptic connection
Typical applications • Clarification of lysates and cell cultures • Concentration, diafiltration, and purification of monoclonal antibodies, plasmids, proteins, viruses, vaccines, colloids, and plasma
GE Healthcare QuixStand benchtop system The QuixStand benchtop system is a compact, easy to use laboratory-scale separations system that uses Xampler cartridges for quick and efficient concentration and diafiltration of a wide range of biological solutions. The QuixStand system is capable of rapidly processing solution volumes up to 10 liters. QuixStand systems can be purchased with or without pumps, and are available in non-sanitary, sanitary, and autoclavable versions.
• Manufacturing material for early stage clinical trials • Scale-up/scale-down studies
GE Healthcare hollow fiber membrane The outstanding performance of GE Healthcare hollow fiber products is based on a proprietary manufacturing process that yields membrane with a structure free of macrovoids and pinhole defects associated with conventional hollow fiber membranes. Separation processes which incorporate a defect-free membrane can use broader operating parameters without compromising membrane performance or integrity.
