Growth Promo on Test Microbiology To lay down a procedure to check the growth promo on proper es of the procured media & also to describes the procedure to be followed to evidence each lot of prepared or autoclaved media the growth of microorganisms when all the environmental condi ons for growth is provided. 1.0 OBJECTIVE: To lay down a procedure to check the growth promo on proper es of the procured media & also to describes the procedure to be followed to evidence each lot of prepared or autoclaved media the growth of microorganisms when all the environmental condi ons for growth is provided. 2.0 SCOPE: The SOP is applicable to the media used for microbiological analysis in microbiology laboratory. 3.0 RESPONSIBILITY: Microbiologist : To perform the procedure as per SOP. Head – Quality Control : To ensure the compliance of the SOP. 4.0 PROCEDURE: 4.1 Liquid broth medium (Growth Promo on / Posi ve Control): 4.1.1 Prepare culture dilu on as per Growth Promo on Test 4.1.2 Inoculate 1 ml of culture to the sterile culture media and incubate at recommended temperature specified for the organism as per Table -1. 4.1.3 The media complies the growth promo on test if luxuriant growth (turbidity) is observed in the inoculated tubes of liquid media or dis nct colony forma on on the agar media within 48 hours of incuba on. 4.1.4 If the observa ons are without any microbial growth discard the batch of medium and reject the en re lot of media.. 4.1.5 Observe inoculated media & enter 4.3 Acceptance criteria :

4.3.1 Micro-organism must show turbidity in the inoculated broth media and dis nct colony forma on on the solid agar media within 48 hours dura on. Table- 1(Rou nely used media )
